This assay (primers and probes only) is used to detect the V600E mutation of the proto-oncogene B-Raf (B-Raf Proto-Oncogene, Serine/Threonine Kinase, BRAF), which encodes a serine/threonine kinase (COSM476, c.1799T>A).
Mutations in the BRAF gene are associated with various cancers, including non-Hodgkin lymphoma, colorectal cancer, malignant melanoma, thyroid cancer, non-small cell lung cancer, and lung adenocarcinoma.
The valine at position 600 in exon 15 of the BRAF gene is replaced by glutamic acid (V600E). This mutation leads to continuous activation of the downstream MEK-ERK signaling pathway, affecting cell division, differentiation, secretion, and crucially influencing tumor growth, proliferation, invasion, and metastasis. Currently, small molecule inhibitors such as vemurafenib and dabrafenib have shown promising inhibitory effects on BRAF V600E.
