This kit is designed for the detection of PIK3CA gene mutations in the DNA of patients with breast cancer, colorectal cancer, and other related conditions. Mutations in the PIK3CA gene can potentially lead to resistance to certain drugs, such as the HER2-targeting trastuzumab and the EGFR-targeting cetuximab monoclonal antibody. The PI3K protein, encoded by the PIK3CA gene, is situated downstream of these monoclonal antibody targets. When PI3K is continuously activated, it remains unaffected by the targeted transmembrane proteins of these monoclonal antibodies.
However, downstream of PI3K, there is also the mTOR protein. Consequently, many studies suggest that drugs targeting the PI3K-AKT-mTOR pathway can be combined with other medications to inhibit tumor growth.
The reagents included in this kit allow for the detection of specific sites associated with PIK3CA gene mutations: C420R, E542K, E545A, E545D, E545G, E545K, Q546E, Q546R, H1047L, H1047R, and H1047Y.
